This package includes50 nmol of SNAP-Surface 649 substrate, sufficientto make 10 ml of a 5 μM SNAP-tag fusion proteinlabeling solution. The SNAP-tag is a novel tool for protein research, allowing the specific, covalent attachment ofvirtually any molecule to a protein of interest. TheSNAP-tag is based on mammalian O6-alkylguanine-DNA-alkyltransferase (AGT). SNAP-tagsubstrates are derivatives of benzylguanines andbenzylchloropyrimidines. In the labeling reaction, the substituted benzyl group of the substrate iscovalently attached to the SNAP-tag.There are two steps to using this system: subcloningand expression of the protein of interestas a SNAP-tag fusion, and labeling of the fusionwith the SNAP-tag substrate of choice. Expressionof SNAP-tag fusion proteins is described in thedocumentation supplied with SNAP-tag plasmids. The labeling of the fusion proteins with the SNAPtagsubstrate is described below.Excitation (dotted line) and emission spectraof SNAP-Surface 649 coupled to SNAP-tag in buffer atpH 7.5Structure of SNAP-Surface 649 (MW 1221.3 g/mol).
