Backgroundinformation
CD326,alsoknownashumanepithelialantigen(HEA),epithelialcelladhesionmolecule(EpCAM),orepithelial-specificantigen(ESA)expressionisseeninthemajorityofcarcinomas.Itexpressedoncellsofepithelialorigin,circulatingtumorcells,andcancerstemcells.
Detailedseparationprocedure
First,cellsarelabeledwithCD326(EpCAM)MicroBeadsandenrichedonthefirst
MACSColumn.Secondly,CD326
+cellsarefixed,appliedtoasecondMACSColumn,andsubjectedtointracellularstainingofthecytokeratinfilamentsusingtwoalkalinephosphatase–conjugatedcytokeratin-specificantibodies,clonesCK3-11D5andCK3-3E4.Afterelutionfromthecolumn,cellsaretransferredontoamicroscopeslide,incubatedwiththesubstrate,andthenanalyzedbylightmicroscopy.Duringtheentirestainingprocedure,thecellsremainretainedwithinthecolumn,thusminimizingcellloss.
