AntarcticPhosphatase(AnP)isaheatlABIlealkalinephosphatasepurifiedfromarecombinantsource.AnPnonspecificallycatalyzesthedephosphorylationof5´and3´endsofDNAandRNAphosphomonoesters.Also,AnPHydrolysesribo-,aswellasdeoxyribonucleosidetriphosphates(NTPsanddNTPs).AnPisusefulinmanymolecularBIOLOGyapplicationssuchastheremovalofphosphorylatedendsofDNAandRNAforsubsequentuseincloningorend-labelingofprobes.Incloning,dephosphorylationpreventsreligationoflinearizedplasmidDNA.Theenzymeactson5´protruding,5´recessed,andbluntends.AnPmayalsobeusedtodegradeunincorporateddNTPsinPCRreactionstopreparetemplatesforDNAsequencingorSNPanalysis.AnPiscompletelyandirreversIBLyinactivatedbyheatingat80°Cfor2minutes,therebymakingremovalofAnPpriortoligationorend-labelingunnecessary.
