ProductBackground:TheS1Pcoated96-wellmicrotiterisblockedtoreducenon-specificbinding.TheuserthenmixestheS1Pstandardcurveandsampleswiththeanti-S1PantibodybeforeaddingthemixturetotheblockedS1Pcoatedplate.Theanti-S1PantibodybindstotheS1PcoatedplateortheS1Pinthesample.Followinganincubationandplatewash,streptavidin-HRPisaddedtotheplateandbindstoallanti-S1Pantibodies(labeledwithbiotin)boundtotheplate.Afteranadditionalincubationandplatewash,TMBsubstrateisaddedtotheplateandthecolorometricreactionstoppedbytheadditionof1Nsulfuricacid.Theabsorbanceat450nmismeasuredandtheconcentrationofS1Pinthesamplesaredeterminedbycomparisontothestandardcurve.
