InthepMAL™ProteinFusionandPurificationSystem,theclonedgeneisinsertedintoapMAL vectordown-streamfromthemalEgene,whichencodesmaltose-bindingprotein(MBP).ThisresultsintheexpressionofanMBP-fusionprotein(1,2,3).ThetechniqueusesthestrongP_tacpromoterandthetranslationinitiationsignalsofMBPtoexpresslargeamountsofthefusionprotein.Thefusionproteinisthenpurifiedbyaone-stepaffinitypurificationspecificforMBP(Figure1)(4).

ThesystemusesthepMALvectorswhicharedesignedsothatinsertionofatargetgeneresultsinanMBPfusionprotein.pMAL-c5XhasanexactdeletionofthemalEsignalsequence,resultingincytoplasmicexpressionofthefusionprotein.pMAL-p5XcontainsthenormalmalEsignalsequence,whichdirectsthefusionproteinthroughthecytoplasmicmembrane.pMAL-p5fusionproteinscapableofbeingexportedcanbepurifiedfromtheperiplasm.BetweenthemalEsequenceandthepolylinkerthereisaspacersequencecodingfor10asparagineresidues.ThisspacerinsulatesMBPfromtheproteinofinterest,increasingthechancesthataparticularfusionwillbindtightlytotheamyloseresin. ThevectorsalsoincludeasequencecodingfortheFactorXarecognitionsite. ThisallowstheproteinofinteresttobecleavedfromMBPafterpurification,withoutaddinganyvector-derivedresiduestotheprotein(5).Forthispurpose,thepolylinkerincludesanXmnIsitesuperimposedonthesequencecodingfortheFactorXacleavagesite,wherethegeneofinterestisinserted.Anumberofotherusefulsitesarepresentdirectlydownstreamtofacilitatecloning.
