GeneralPurposeReagents
PrincipleofProcedure:
TheIHCSelect®ImmunoperoxidaseSecondaryDetectionSystemusesbiotin-avidin-HRPcomplexedantibodiestodetectrabbitormouseIgGinthePrimaryAntibody.Theprimaryantibodybindstospecificantigenspresentinthespecimen.Anyexcessantibodyisremovedbywashing.Next,theSecondaryAntibodiesReagent(conjugatedtobiotin)isaddedandwillreactwiththeprimaryantibodypresentonthetissue.Unboundantibodyisremovedbywashing.ThesampleisthenbyincubatedwiththeStreptavidin-HRPsolution,whichbindstothebiotin-labeledsecondaryantibodypresentonthetissue.Unboundenzymeisremovedbywashing.Thechromogenicdevelopmentreagent,3,3"diaminobenzidine(DABSubstrate),isthenaddedandreactswiththeHRPattachedtotheHRP-streptavidin-biotin-antibodycomplex.TheHRPactivityonthechromogenicsubstrateresultsinthedepositofbrowntoblackinsolubleprecipitateatthoseantigenicsitescontainingthespecificepitopesrecognizedbytheprimaryantibody.Tissuesprocessedinavarietyoffixativesincludingneutralbufferedformalin,B-5,andBouin"smaybeused.
