Ac-Arg-Gly-Phe-Phe-Pro-AFC is a fluorogenic substrate for cathepsin D. Cathepsin D is a lysosomal aspartyl protease whose role is to degrade and activate intracellular proteins including hormones, enzyme precursors, and growth factors. Over expression in tumors stimulates metastasis and considered a marker of poor prognosis in breast cancer.
Activity is quantified by release of free fluorescent 7-amino-4-trifluoromethylcoumarin (AFC) which excites at 395-400 nm and emits at 495-505 nm.
