Ac-Val-Glu-Ile-Asp-pNA is a colorimetric substrate for Caspase 6. Caspase 6 (Mch2) is a cysteine protease involved in the execution phase of apoptosis, mediating neuronal shrinkage. In addition, it has important roles in neurodegenerative disorders such as Huntington’s, Parkinson’s, and Alzheimer’s diseases. Caspase 6 cleaves the mutated huntintin protein and the amyloid precursor protein resulting in the aggregating protein fragments typical of Huntington’s and Alzheimer’s diseases respectively.
Activity is quantified by the release of p-nitroaniline which is measured by absorbance at 405 nm.
