Caspase 1 (ICE) Substrate (Ac-Tyr-Val-Ala-Asp-AMC) is a fluorogenic substrate for interleukin 1b converting enzyme (ICE) which is a cysteine protease that cleaves specifically at aspartic acid. ICE converts inactive IL-1 beta precursor to a pro-inflammatory cytokine that mediates a wide range of immune and inflammatory response. ICE-related cysteine proteases are required for E1A-induced, p53-dependent apoptosis in baby rat kidney (BRK) cells. Adenovirus E1B 19K protein, which is a potent inhibitor of apoptosis, inhibits activation of these proteases in BRK cells.
Activity is quantified by release of free fluorescent 7-amino-4-methylcoumarin (AMC) which excites at 360-380 nm and emits at 440-460 nm.
