Furin inhibitor II (Hexa-D-Arginine), as the name implies, inhibits furin (Ki = 106 nM for purified furin). Furin is an enzyme belonging to the subtilisin-like proprotein convertase family which process precursor proteins into biologically active products.Representative substrates include von Willebrand factor, TGFβ-1 precursor, MMP-1, gp160, proparathyroid hormone and proalbumin. Results from a recent paper and preprint show the presence of a furin cleavage site on the spike protein of the SARS-CoV-2 which is required for membrane fusion and replication. Inhibition of furin activity can stop viral replication.
