ITS (Insulin, Transferrin, Selenium) is a chemically definedsupplement for a variety of cell culture applications. ITSsupplement contains components which are required for optimalperformance of serum-free media. Supplementation of ITS todifferent conventional nutrient media substantially reduces theFBS requirement for routine maintenance of many cell lines.
Components: Contains 1.0 mg/ml insulin from bovine pancreas, 0.55mg/ml human transferrin (substantially iron-free), and 0.5 μg/mlsodium selenite.
ROLE OF THE COMPONENTS
INSULIN is a polypeptide hormone that promotes the uptake ofglucose and amino acids and may owe observed mitogenic effect tothis property.
TRANSFERRIN is an iron-transport protein. Iron is an essentialtrace element, but can be toxic in the free form. To nourishcells in culture, it is supplied bound to transferrin inserum.
SELENIUM is an essential trace element normally provided byserum.
Nutritional studies indicate that the supplement componentsdescribed are utilized by most mammalian cells. They enhance cellproliferation and decrease the serum requirement of many celltypes. When the provided supplement is used with 2 to 4 percentserum, proliferation is reported to be similar to mediumsupplemented with 10 percent serum.
