Dynabeads®M-270Epoxybeadsare2.8µmsuperparamagneticbeadscontainingsurfaceepoxygroups.Thebeadscovalentlybindprimaryaminoandsulfhydrylgroupsinproteinsandpeptides,makingthemidealforcouplingantibodies,peptides,intactproteins,andfunctionalenzymes.Thesehydrophylic,neutralpHbeadsexhibitextremelylownonspecificbindingofproteinsanddyes,whichreducestheneedforblockingagents.Also,Dynabeads®M-270EpoxyBeadsdonotcontainTween®detergent,andsoareidealforuseinmassspectrometry(MS)analysis.

CouplingprocedureCovalentcouplingisperformedovernightbyincubatingthedesiredligand(commonlyantibodies,peptides,orproteins)withDynabeads®M-270Epoxybeads.Thebindingcapacityvariesdependingontheligandused,butistypically5–10µgIgGpermilligramofbeads.CouplingoccursatneutralpHandhighsaltconcentration.Thecouplingreactioncanbeperformedoverabroadrangeoftemperaturestomatchthestabilitycharacteristicsofyourligand.
Uponcompletionoftheligandcouplingstep,thereactivemoietiesontheDynabeads®M-270Epoxybeadsaredeactivatedtopreventfurthercoupling.Oncethecouplingreactioniscompleted,thesenonporous,hydrophilic,pHneutralbeadsexhibitextremelylownonspecificbinding.
