StemMACS™DAPTisacell-permeabledipeptidethatinhibitstheproteaseγ-secretase,akeycomponentoftheNotchsignalingpathway.TheNotchpathwayisinvolvedinvariousdevelopmentalcellfateandlineagedecisions,makingthesmallmoleculeinhibitorDAPTavaluabletoolforthemodulationofstemcelldifferentiationincellculture.Inaddition,γ-secretaseplaysaroleinprocessingofamyloidprecursorprotein(APP)toβ-amyloid,acomponentofthesenileplaquescharacteristicforAlzheimer’sdisease.Inhibitionofγ-secretasebyDAPThasbeenshowntocauseadose-dependentreductionofβ-amyloidlevelsinprimaryneuronalculturesaswellasinvivo.
