Ciliaryneurotrophicfactor(CNTF),alsoknownasmembrane-associatedneurotransmitterstimulatingfactor(MANS),isapotentneuralfactorthatwasinitiallypurifiedfromchickembryooculartissue.IntermsofneuronalspecificityCNTFoverlapswithneurotrophins,althoughitscellularlocalisation,receptorstructure,andsignalingpathwayaredistinctfromthoseoftheneurotrophinfamily.TheCNTFαreceptorhasbeenlocalizedtoskeletalmusclesandtothenervoussystem.Onereceptorsubunitisknownasgp130whichisalsoacomponentofreceptorsfortheleukaemiainhibitoryfactor(LIF),interleukin6(IL-6),interleukin11(IL-11),andoncostatinM(OSM).CNTFispresentinnormalperipheralnerves,localisedmainlyinthecytoplasmofmyelin-associatedSchwanncellsaswellasastrocytesinthecentralnervoussystem.Itpromotesthesurvivalofneuronsandoligodendrocytesaswellassensoryneurons,motorneurons,basalforebrainneurons,andtype2astrocytes.CNTFishighlyconservedacrossspeciesandexhibitscross-speciesactivity.
