Backgroundinformation
CD146isatransmembraneglycoproteinandbelongstotheIgGsuperfamilyofcelladhesionmolecules.1TheCD146(LSEC)antibodybindstomouseliversinusoidalendothelialcells(LSECs)whicharemicrovascularendothelialcellsliningthehepaticsinusoidalwallandmainlycontributetothecontrolofimmuneresponsesagainstcirculatingsolubleantigensintheliver.Theirstrategicpositioningfavorsatightinteractionwithlymphocytesmigratingthroughtheliver.LSECspossessahighcapacityforantigenuptakeandprocessingbutexpress,incontrasttoprofessionalantigen-presentingcells(e.g.dendriticcells),onlylowlevelsofcostimulatorymolecules.2
TheCD146(LSEC)antibodyalsobindstoendothelialcellsfromavariousrangeoforgans,suchasskin,liver,kidney,brain,spleen,lymphnode,intestine,heartandskeletalmuscleaswellasonbloodvesselstructures,suchaspulmonaryarteries,veins,andthecapillarynetworkofthealveolarwallsbutnotonlymphaticendothelium.IncontrasttohumansthatexpressCD146alsoonTcellsorfolliculardendriticcells,murineCD146expressionwasonlyfoundonasubsetofNK1.1+cellsatlowlevels.3
