  Backgroundinformation
  ThematrixoftheLargeCellColumnsiscomposedofferromagneticspheres,whicharecoveredwithacell-friendlycoatingallowingfastandgentleseparationofcells.WhenplacedinthemagneticfieldofaMACS^®Separator,thespheresamplifythemagneticfieldby10,000-fold,thusinducingahighgrADIentwithinthecolumn.ThisiscrucialforisolationofcellswhichareonlyminimallylabeledwithMACS^®MicroBeads,leavingenoughepitopesfreeforconcurrentantibodystaining.Thespacebetweenthespheresisseveraltimeslargerthanprimaryandmostculturedcells.Thisallowsthecellstofreelyflowthroughthecolumn.Magneticallylabeledcellsareheldinsuspensionwithinthecolumnanddonotactually“bind”thecolumnmatrix.Thissuspensionminimizesstressonthecellsandallowsforefficientsterilewashingbyavoidingcellaggregation.
